Sampling Design
Soil and sediment samples were collecting using standard CALeDNA pre-labeled tubes. Sediment samples were collected approximately 10m apart along an estimated 100m transect. In areas where the river was too deep to collect sediment from the bottom, sediment samples were collected from the banks of the river.
Water samples were collected using two methods:
- plankton net (PN): an adaptor with a cellulose nitrate filter is tied to the end of a plankton net. The net is then dragged through the river for an average of 10-15m
- drip bag (DB): a 1L Nalgene bottle was used to collect a set amount of water that is then placed into an IV bag. These bags are stored in the cold room on campus and rely on gravity to drain into a cellulose nitrate filter overnight.
